If I take my youth team, they all lost more fitness on 29 August than on 27 and 31.
In the case of one of my full-backs, 46% on the 27th, 52% on the 29th and 46% on the 31st. I see the same trend for all the players who played on those 3 days.
If anything, your players will lose less physicality today than they did in the two previous games.

Si je prends mon équipe de jeunes, ils ont tous perdus plus de physique le 29 août que le 27 et le 31.
Pour le cas d'un de mes latéraux, 46% le 27 ; 52% le 29 ; 46% le 31. J'observe la même tendance sur tous les joueurs qui ont joué ces 3 jours là .
Si ça se trouve aujourd'hui tes joueurs vont perdre moins de physique que les deux matchs précédents.
I can totally confirm this, as I've also seen this trend for several weeks/months now. A factor independent of the instructions used (random or not, or the impact of the opposing instructions) means that some matches result in more physical losses (~5% at most).
